Effect of Bacillus polyfermenticus SCD on antioxidative metabolism in hypercholesterolemic rats

Hypercholesterolemia and formation of oxidized LDL are believed to be important in the etiology of coronary heart disease (CHD). On the other hand, it has been shown that some antioxidants such as carotenoids and flavonoids are beneficial to CHD. Recently, we have found that B. polyfermenticus SCD has antioxidative effect from an in vitro study. The purpose of this study was to evaluate the effect of B. polyfermenticus SCD supplementation on antioxidative metabolism in hypercholesterolemic rats. The 50 of 8 wk-old male Sprague-Dawley rats were divided into three groups after 1 week of adaptation period and fed with high fat (12g/100g)-high cholesterol (1g/100g) diet (Control), high fat-high cholesterol diet supplemented with low- or high-doses (3.1x106 cfu/day or 3.1x108 cfu/day) of B. polyfermenticus SCD for 6 weeks. Total antioxidant capacity was determined using Trolox equivalent antioxidant capacity (TEAC) assay in plasma. We also measured the antioxidant enzyme activities in erythrocytes. Lipid peroxidation was determined by analyzing baseline diene conjugation in LDL lipids (LDL-BDC). The total antioxidant capacity of plasma increased significantly in B. polyfermenticus SCD supplemented groups. However, the antioxidant enzymes in erythrocytes and plasma lipid peroxidation levels did not correlate with the B. polyfermenticus SCD supplementation. These results suggest that B. polyfermenticus SCD can increase the plasma antioxidant capacity and thus improve oxidative stress in hypercholesterolemic rats.
