Comparative study for dedection of B-group vitamins and folic acid by gas chromatography-mass spectrometry (GC-MS) and differential pulse polarography (DPP) in sweetpotato (Ipomea Batatas L.) |

The need for regular monitoring of vitamins in foods and food products has led to an increasing demand for adequately selective analytical techniques with sensitive determination capabilities. Differential Pulse Polarography (DPP) has become accepted as one of the most powerful voltammetric electroanalytical tool for minor components in foods and biological matrices due to its extreme sensitivity and selectivity. Sweetpotato (Ipomea Batatas L.) genotype named “Hatay Red” with orange flesh color was used as genetic material and cultured in vitro ,then planted at the Aegean University Department of Field Crops. New plant “Hatay Red Bornova” was harvested in November of 2002 and dry-milled into flours prior electroanalytical and GC-MS analysis. A polarographic analyzer (METROHM 746 VA Trace Analyzer) together with a capillary hanging mercury drop electrode (HMDE) was used for all electroanalytical measurements. A platinum wire was used as the counter electrode and Ag/AgCl was the reference electrode. HP 5400/5973 model gas chromatograph using a 30 m, 0.25 mm, 0.25 µm capillary column HP-5-MS (J & W Scientific, Folsom, CA) was used for Gas-Chromatography Mass Spectrometric (GC-MS) analysis. Vitamin B1 (Thiamin), B2 (Riboflavin), B6 (Pyridoxine), B12 (Cyanocobalamine), Niacin, and Folic Acid in this new generated genotype of sweetpotato were performed by using GC-MS and DPP techniques as comparatively. The proposed Differensial Pulse Polarographic (DPP) method is a rapid, sensitive and reproducible for dedection of B-group vitamins and folic acid in food quality control area.
