Effect of prebiotics on azoxymethane(AOM)-induced aberrant crypt foci (ACF) in colon of Fisher 344 rats

Colorectal cancer is the leading cause of cancer deaths in the US. Preboiotics, namely Oligofructose and Inulin, which are selective fermentable fructans have potential chemopreventive activity. The objective of this study was to test the effect of Chicory inulin and oligofructose on azoxymethane (AOM) induced aberrant crypt foci (ACF) in rat colon. After an acclimatization period of one week, 5 groups of Fisher 344 male weanling rats were assigned to AIN 93G (control), AIN 93G containing 5 and 10% levels of oligofructose and inulin, respectively. Rats received 2 s/c injections of AOM in saline at 16mg/kg body wt at 7 & 8 wk of age. The animals continued to receive the assigned diets until sacrificed by CO2 euthanasia at 17 wk of age and colon samples collected for enumeration of ACF. Body weights were comparable in all groups. There was no significant difference (p>0.05) in feed intake between control and experimental groups. There was a significant (p<0.05) increase in cecal weight and a decrease in cecal pH from 7.58 in control group to 7.04, 6.02, 6.40, 5.65, in the groups fed oligofructose and inulin at 5 and 10% levels, respectively. Compared to the control group, the percent reductions of ACF in 5 and 10% oligofructose groups were 20 and 28, and in the 5 and 10% inulin groups, 40 and 46, respectively. The results of this study indicate that dietary oligofructose and inulin which are prebiotics suppress AOM-induced ACF formation, an early preneoplastic marker in the process of colon tumorigenesis in Fisher 344 male rats. The designing of prebiotic based foods maybe of significance in the food industry.
