Genistein reduces UVB induced oxidative damages in cultured human skin fibroblasts

Genistein is well known active component that mainly occurs in soybean and whole-grain products. It seems that health- benefical effects of genistein are related to their hormonal and antioxidant activities. Our objective was to determine UVB protective effects on skin fibroblast cells and related antioxidant activities of genistein. To assess the UVB protective effect of genistein, we treated fibroblasts with varying concentrations of genistein before UVB radiation (200mJ/cm2). Trypan Blue exclusion test and SRB (Sulphorhodamine B) asaay were performed to decide cytotoxicity of genistein on UVB damaged cells. For further analysis of protective effects of genistein treatment on UVB induced apoptosis, annexin V binding and Propidium iodide(PI) uptake was also measured using FACS. Trypan Blue exclusion test showed that genistein treatment resulted in less cell death compared to UVB radiated control group. According to SRB assay, fibroblasts treated with 0.01, 0.1, 1, 10mg/ml of genistein showed significantly increased cell viability with a dose related pattern compared to UVB damaged, untreated cells. FACS study indicated that genistein reduced UVB induced G0/G1 population by 66.91% compared to only UVB radiated cells (78.27%). It was also shown that, in presence of genistein, UVB induced apoptosis were decreased to 8.82% with 10mg/ml, 4.34% with 1mg/ml, 3.3% with 0.1mg/ml, 3.5% with 0.01mg/ml and 3.55% with 0.001mg/ml. This study suggests that genistein inhibits UVB induced apoptosis in skin fibroblasts. It seems that the UVB protective effects of genistein is partly due to their antioxidant action.
